Glycolytic Rate Assay

Glycolytic Rate Assay is a method for quantifying the rate at which living cells convert glucose to pyruvate through glycolysis, providing a functional measure of cellular energy metabolism. The assay typically monitors extracellular acidification, generated largely by lactate and proton release during glycolysis, while mitochondrial respiration is perturbed with pathway-specific inhibitors to distinguish glycolytic from respiratory acidification. In neuroscience, it can compare metabolic phenotypes in neurons, astrocytes, microglia, or neural models under different nutrient, activity, or disease conditions. These measurements help assess metabolic flexibility, identify shifts between glycolysis and oxidative phosphorylation, and evaluate how bioenergetic changes contribute to neural function or dysfunction.

Glycolysis is a defining metabolic marker in multiple biological systems. Monitoring glycolysis by measuring the extracellular flux of H+ is common, but requires correction to be quantitative and unambiguous. Here, we demonstrate how to gather and correct extracellular flux data to distinguish between respiratory and glycolytic sources of extracellular acidification.

The pentose phosphate pathway (PPP) operates in parallel with glycolysis, facilitating the metabolism of both pentoses and glucose. This pathway consists of two distinct phases: the oxidative and non-oxidative phases. While it does not directly generate ATP, the intermediates formed during the process can integrate into glycolysis, contributing to cellular energy metabolism when required.Oxidative Phase: NADPH ProductionThe oxidative phase of the pentose phosphate pathway is primarily...

Here we present two protocols, one for measuring the specific growth rate and the other for the cell-binding ability of rotavirus using the plaque assay and RT-qPCR. These protocols are available for confirming the differences in phenotypes between rotavirus strains.
