Nbd-ph Fapp

NBD-pH FAPP is a biochemical fluorescence-based approach for monitoring acidity in molecular or cellular environments, using an NBD-derived reporter whose signal responds to pH. Its mechanism depends on pH-sensitive changes in the fluorophore’s protonation state, which alter fluorescence intensity or spectral properties and allow measurements after calibration against solutions of known pH. In biochemistry, the method can help evaluate proton gradients, acidic compartments, membrane-associated processes, and pH-dependent molecular interactions. By converting local chemical conditions into a measurable optical signal, NBD-pH FAPP supports quantitative studies of dynamic biochemical systems and can complement microscopy, spectroscopy, and other fluorescence assays.

SNARE-mediated Fusion of Single Proteoliposomes with Tethered Supported Bilayers in a Microfluidic Flow Cell Monitored by Polarized TIRF Microscopy

Here, we present a protocol to detect single, SNARE-mediated fusion events between liposomes and supported bilayers in microfluidic channels using polarized TIRFM, with single molecule sensitivity and ~15 msec time resolution. Lipid and soluble cargo release can be detected simultaneously. Liposome size, lipid diffusivity, and fusion pore properties are measured.

In this video, we demonstrate genetic transformation of the rice genome using an Agrobacterium tumefaciens binary vector. This bacterium facilitates the transfer of a foreign DNA plasmid using Agrobacterium virulence proteins.
