3d Epithelial Tissues

3D epithelial tissues are laboratory-engineered or cell-derived models that reproduce the organization and barrier functions of epithelial layers in three dimensions, making them valuable for bioengineering and biomedical research. Epithelial cells establish apical-basal polarity, form cell-cell junctions, and interact with an extracellular matrix to self-organize into structures such as spheroids, organoids, or layered tissues. These models can capture transport, secretion, tissue remodeling, and responses to chemical or mechanical stimuli more realistically than two-dimensional cultures. Applications include studying development and disease, evaluating drug toxicity, designing tissue-engineered grafts, and developing platforms for regenerative medicine.

Tissue Engineering of a Human 3D in vitro Tumor Test System

Methods to create human 3D tumor tissues as test systems are described. These technologies are based on a decellularized Biological Vascularized Scaffold (BioVaSc), primary human cells and a tumor cell line, which can be cultured under static as well as under dynamic conditions in a flow bioreactor.

Culturing and Applications of Rotating Wall Vessel Bioreactor Derived 3D Epithelial Cell Models

A rotating cell culture system that allows epithelial cells to grow under physiological conditions resulting in 3-D cellular aggregate formation is described. The aggregates generated display in vivo-like characteristics not observed in conventional culture models and serve as a more accurate organotypic model system for a multitude of scientific investigations.

Engineering Three-dimensional Epithelial Tissues Embedded within Extracellular Matrix

This manuscript describes a soft lithography-based technique to engineer uniform arrays of three-dimensional (3D) epithelial tissues of defined geometry surrounded by extracellular matrix. This method is amenable to a wide variety of cell types and experimental contexts and allows for high-throughput screening of identical replicates.

We describe a method to establish human enteroids from small intestinal crypts and colonoids from colon crypts collected from both surgical tissue and biopsies. In this methodological article, we present the culture modalities that are essential for the successful growth and maintenance of human enteroids and colonoids.
