4d Printing

4D printing is an extension of additive manufacturing that produces three-dimensional structures designed to change shape or function over time, making it valuable for engineering dynamic biological systems. The process uses programmable, stimuli-responsive materials, such as shape-memory polymers or hydrogels, whose properties respond to conditions including heat, moisture, pH, or mechanical forces after fabrication. In bioengineering, these transformations can create tissue-engineering scaffolds that adapt to their surroundings, drug-delivery systems with controlled release, and devices that deploy inside the body with minimal intervention. By linking material design, fabrication, and biological function, 4D printing supports the development of more adaptive implants and regenerative therapies.

Using a 3D printer, a shape memory polymer filament is extruded to form a branched tubular structure. The structure is patterned and shaped such that it can contract into a compact form once folded and then return to its formed shape when heated.

This protocol describes the analysis of fluorescently labeled intracellular compartments in budding yeast using multi-color 4D (time-lapse 3D) confocal microscopy. The imaging parameters are chosen to capture adequate signals while limiting photodamage. Custom ImageJ plugins allow labeled structures to be tracked and quantitatively analyzed.
