Endothelial Cell Viability

Endothelial cell viability is the capacity of endothelial cells to remain alive, metabolically active, and functionally intact under defined conditions. It is commonly evaluated by measuring membrane integrity, metabolic activity, and responses to stress, because cell death or dysfunction can compromise barrier formation and vascular signaling. In bioengineering, viability assessment helps optimize culture conditions, biomaterials, microfluidic vessels, and tissue-engineered vascular grafts; viable endothelial layers can support selective permeability, reduce the thrombogenicity of surfaces, and improve the physiological relevance of engineered vascular models.

Viability Assays for Cells in Culture

Therapeutic compounds are often first examined in vitro with viability assays. Blind cell counts by a human observer can be highly sensitive to small changes in cell number but do not assess function. Computerized viability assays, as described here, can assess both structure and function in an objective manner.

Determining the number of cells in culture is important in standardization of culture conditions and in performing accurate quantitation experiments. In this video, we demonstrate how cells are counted using a hemacytometer.

Isolation of Murine Embryonic Hemogenic Endothelial Cells

Hematopoietic stem and progenitor cells (HSPC) derive from specialized (hemogenic) endothelial cells during development, yet little is known about the process by which some endothelial cells specify to become blood forming. We demonstrate a flow-cytometry based method allowing simultaneous isolation of hemogenic endothelial cells and HSPC from murine embryonic tissues.

We provide a method for isolating and culturing pure populations of heart valve endothelial cells (VEC). VEC can be isolated from either side of the cusp or leaflet and immediately following, underlying interstitial cell (VIC) isolation is straightforward.
