Epithelial Marker

An epithelial marker is a gene, protein, or surface molecule used to identify epithelial cells and assess their state, making it important for characterizing engineered tissues and cell-based models. Common markers such as cytokeratins, E-cadherin, and EpCAM reflect epithelial identity through intermediate-filament networks, adherens junctions, or cell-surface expression; changes in their abundance or localization can indicate differentiation, barrier formation, or epithelial-to-mesenchymal transition. In bioengineering, immunostaining, flow cytometry, and gene-expression assays use these markers to verify tissue organization, monitor organoid and scaffold-based cultures, and evaluate how materials or mechanical cues influence epithelial function and stability.

An example of adenoviral gene therapy in the human diabetic organ-cultured corneas is presented towards the normalization of delayed wound healing and markedly reduced epithelial stem cell marker expression in these corneas. It also describes the optimization of this process in stem cell-enriched limbal epithelial cultures.

The fallopian tube (FT) is emerging as an alternative site of origin for serous ovarian carcinoma (SOC). This protocol describes a novel method for the isolation and ex vivo culture of fallopian tube epithelial cells. This system recapitulates the in vivo epithelium and allows the study of SOC pathogenesis.
