Point Spread Function Calibration

Point spread function calibration is the measurement and characterization of how an imaging system represents a point-like object, a critical step for determining resolution and correcting optical distortions. In bioengineering, calibration commonly uses sub-resolution fluorescent beads to record the system’s three-dimensional intensity distribution, or point spread function, under defined imaging conditions. The measured function can reveal changes in focus, aberrations, alignment, and performance across spatial dimensions or experimental settings. These data support quantitative microscopy, image deconvolution, and reliable measurements of cellular structures, biomaterials, and engineered tissues, while enabling researchers to compare instruments and validate imaging workflows.

Source: Laboratory of Dr. B. Jill Venton - University of Virginia Calibration curves are used to understand the instrumental response to an analyte and predict the concentration in an unknown sample. Generally, a set of standard samples are made at various concentrations with a range than includes the unknown of interest and the instrumental response at each concentration is recorded. For more accuracy and to understand the error, the response at each concentration can be repeated so an error...

Spreading of Chromatin Modifications

The histone proteins in the nucleosomes are post-translationally modified (PTM) to increase or decrease access to DNA. The commonly observed PTMs are methylation, acetylation, phosphorylation, and ubiquitination of lysine amino acids in the histone H3 tail region. These histone modifications have specific meaning for the cell. Hence, they are called "histone code". The protein complex involved in histone modification is termed as "reader-writer" complex. Writers The writer is an enzyme that can...

Accurate calibration of glassware, such as volumetric flasks, pipettes, and burettes, is essential to ensure accurate measurements in the analytical laboratory. Calibration helps maintain consistency across measurements and prevents errors arising from inaccurate volumes. Volumetric flasks: Volumetric flasks are designed to prepare aqueous solutions of precise volumes accurately with a calibration line on the neck. To calibrate a volumetric flask, it is important to fill it with distilled...
