Plant Tissue Analysis

Plant tissue analysis is the systematic examination of plant cells, tissues, and their chemical or structural characteristics to understand growth, development, health, and responses to environmental conditions. It typically involves collecting representative tissue samples, preserving or homogenizing them, and applying methods such as microscopy, staining, biochemical assays, or molecular analysis to measure cellular structures, metabolites, proteins, or nucleic acids. In biological research, these approaches help identify nutrient deficiencies, pathogen effects, stress responses, and developmental changes. Plant tissue analysis also supports crop improvement, physiology studies, biotechnology, and the evaluation of how plants respond to factors such as light, water availability, temperature, and soil composition.

Robotics and Dynamic Image Analysis for Studies of Gene Expression in Plant Tissues

We report a method for introduction, tracking and quantitative analysis of GFP expression in plant cells. This method utilizes a custom-designed robotics system for semi-continuous image collection from large numbers of samples, over time. We also demonstrate the use of ImageJ and ImageReady for analysis of image series.

This article presents a method for the detection and quantification of organic acids from plant material using free zonal capillary electrophoresis. An example of the potential application of this method, determining the effects of a secondary fermentation on organic acid levels in coffee seeds, is provided.

Here we describe a new method to study protein import into isolated chloroplasts under stress. The method is rapid and straightforward, and can be applied to study the consequences of different stress conditions for chloroplast protein import, and the corresponding regulatory mechanisms.

Immunolocalization of Arabinogalactan Proteins and Pectins in Plant Tissue

This video demonstrates an immunostaining technique to localize arabinogalactan proteins (AGPs) and pectins in plant tissues. Fixed and resin-embedded plant tissue sections are stained with AGP- and pectin-specific primary and fluorescent secondary antibodies and a cellulose-binding fluorescent dye to reveal their tissue-specific colocalization in the cell wall.
