Aphid Microinjection

Aphid microinjection is a specialized laboratory technique for introducing small volumes of solutions into aphids or their embryos, enabling direct experimental manipulation of these small-bodied insects. Under a microscope, researchers immobilize the insect and use a pulled glass capillary to penetrate a selected region, delivering materials such as nucleic acids, dyes, drugs, or microbial suspensions while minimizing tissue damage and desiccation. The method supports studies of aphid development, physiology, host interactions, symbiosis, and gene function, and can complement RNA interference or genetic manipulation approaches. By enabling controlled delivery at defined stages, it helps connect molecular mechanisms with organism-level phenotypes.

Aphids are effective transmitters of plant viruses. Aphid microinjection of virus, the procedure we will show you today, is a technique allowing researchers to inject virus directly into the hemocoel of the aphid, bypassing the gut, one of the 2 major barriers for virus transmission in a circulative manner. The same technique is also used to inject dsRNA for RNAi.
