Cartilage Matrix Analysis

Cartilage matrix analysis is the study of the composition, organization, and condition of the extracellular matrix that gives cartilage its strength, resilience, and load-bearing capacity. It combines approaches such as histological staining, microscopy, and biochemical assays to assess matrix components, including collagen and proteoglycans, as well as their spatial distribution and abundance. In biology, these measurements help characterize cartilage development, tissue maintenance, degeneration, and repair by revealing changes in matrix structure and content. The resulting data support research on joint disease, biomaterials, tissue engineering, and regenerative strategies aimed at restoring functional cartilage.

This paper introduces a new method for the synthesis of decellularized cartilage extracellular matrix (DC-ECM) hydrogels. DC-ECM hydrogels have excellent biocompatibility and provide a superior microenvironment for cell growth. Therefore, they can be ideal cell scaffolds and biological delivery systems.

Cell organization of craniofacial bones has long been hypothesized but never directly visualized. Multi-spectral cell labeling and in vivo live imaging allows visualization of dynamic cell behavior in zebrafish lower jaw. Here, we detail the protocol to manipulate Zebrabow transgenic fish and directly observe cell intercalation and morphological changes of chondrocytes in the Meckel’s cartilage.
