Caspase Activation

Caspase activation is the process by which inactive cysteine proteases become active to regulate programmed cell death and inflammatory responses. In apoptosis, cellular stress or death-receptor signaling activates initiator caspases, which cleave and activate executioner caspases; these enzymes then dismantle the cell by targeting structural and regulatory proteins. Other caspases can process inflammatory mediators and contribute to innate immune defense. Because activation occurs through tightly controlled proteolytic cascades, it helps maintain tissue balance while limiting damage to neighboring cells. Studying caspase activation supports research on development, cancer, neurodegeneration, immune disorders, and the evaluation of drugs that alter cell survival.

In Vivo Biosensor Tracks Non-apoptotic Caspase Activity in Drosophila

To detect healthy cells in whole animals that contain low levels of caspase activity, the highly sensitive biosensor designated CaspaseTracker was generated for Drosophila. Caspase-dependent biosensor activity is detected in long-lived healthy cells throughout the internal organs of adult animals reared under optimized conditions in the absence of death stimuli.

Visualization of Caspase Activation in Human Monocyte-Derived Macrophages

This video demonstrates the activation of caspase proteins in human macrophages using bimolecular fluorescence complementation reporter proteins. The pro-domains of caspase-1 are fused with non-fluorescent fragments of Venus proteins, which are recruited to inflammasome complexes. The proximity of inflammasomes induces the refolding and fluorescence of Venus fragments, confirming caspase activation in the macrophages.

Certain genetic perturbations or exposure to toxins can disrupt normal developmental processes leading to death of specific cell types. The analysis of activated Caspase 3 by whole-mount immunofluorescence in zebrafish embryos reveals stage- and tissue-specific localization of cells specifically undergoing apoptosis.
