Gus Activity Measurement

GUS activity measurement is a method for quantifying the activity of β-glucuronidase, an enzyme commonly used as a reporter of gene expression in biological research. In this assay, β-glucuronidase cleaves glucuronide-linked substrates such as 4-methylumbelliferyl-β-D-glucuronide or X-Gluc, producing a fluorescent or colored product whose signal reflects enzyme activity. Measurements can therefore reveal when and where a promoter is active, enabling analysis of gene regulation, tissue-specific expression, developmental patterns, and transformation efficiency in plants and other experimental systems. Quantitative assays support comparisons across samples, while histochemical staining provides spatial information within cells or tissues.

Programmed cell death assays commonly used in mammalian systems such as DNA laddering or TUNEL assays, are often difficult to reproduce in plants. In combination with a GUS reporter system, we propose a rapid, plant based transient assay to analyze the potential death properties of specific genes.

Measurement of Larval Activity in the Drosophila Activity Monitor

This report describes a method for measuring Drosophila larval activity using the TriKinetics Drosophila Activity Monitor. The device employs infrared beams to detect movements of up to 16 individual animals. Data can be analyzed to represent motion parameters including rates and the positions of the animals within the assay chambers.

Here, we present two protocols to transform potato plants. The Agrobacterium tumefaciens transformation leads to a complete transgenic plant while the Agrobacterium rhizogenes produces transgenic hairy roots in a wild type shoot that can be self-propagated. We then detect promoter activity by GUS staining in the transformed roots.
