Cdna Circularization

cDNA circularization is a molecular biology technique that converts linear complementary DNA (cDNA) into a covalently closed circular molecule, enabling specialized analysis of transcript sequences. The process brings the cDNA ends together and joins them through enzymatic ligation, often using a complementary splint or end-compatible design to promote accurate circular formation. Circular cDNA can resist degradation by exonucleases and serve as a template for repeated copying, including rolling circle amplification, which generates linked copies of the original sequence. These properties support applications such as transcriptome analysis, isoform characterization, mutation detection, and highly sensitive sequencing workflows.

The cDNA microarray PtGen2 was developed for gene expression studies in loblolly pine, P. taeda, and other conifer species. Here, we show pre- and post-hybridization handling and washing techniques that can be used with this array to yield better consistency, reduced artifacts, and lower backgrounds.
