Lucifer Yellow Dye

Lucifer Yellow dye is a water-soluble fluorescent tracer used to visualize individual cells, their processes, and connections in biological tissues. Because the dye is generally membrane-impermeant, researchers introduce it into selected cells by iontophoretic injection or through a patch pipette, where its fluorescence reveals cytoplasmic structure under a fluorescence microscope. Lucifer Yellow can also pass through functional gap junctions, allowing investigators to assess intercellular coupling and communication. In biology, it supports neuronal tracing, cell morphology studies, developmental research, and analysis of tissue organization, providing a practical way to link cellular structure with connectivity and physiological interactions.

Yellow Fever

Yellow fever is a viral hemorrhagic disease caused by the yellow fever virus (YFV), a member of the Flaviviridae family. It is transmitted primarily by Aedes and Haemagogus mosquitoes in tropical and subtropical regions of Africa and South America. After transmission through a mosquito bite, the virus initially replicates in skin-resident immune cells such as dendritic cells and macrophages. These cells then migrate to the lymph nodes, where viral replication increases, eventually leading to...

Source: Chang, X., et al. Preparation of Pseudo-Typed H5 Avian Influenza Viruses with Calcium Phosphate Transfection Method and Measurement of Antibody Neutralizing Activity. J. Vis. Exp. (2021).This video demonstrates a luciferase-based assay to quantify pseudovirus infection in epithelial cells. The pseudovirus encodes the luciferase gene, which is expressed in infected cells. After washing, and cells are incubated with lysis buffer at an ultra-low temperature, and then thawed to induce lysis.

Transfection into the macrophage cell line, RAW264.7, is difficult due to the cell’s natural response against foreign materials. We described here a gentle yet robust procedure for transfecting luciferase reporter genes into RAW264.7 cells.
