Mosquito Toxicity Assay

A mosquito toxicity assay is a laboratory method for measuring how a chemical, biological agent, or environmental substance affects mosquito survival and behavior, making it important for evaluating vector-control strategies. In a typical assay, mosquitoes receive a controlled exposure to a test substance, and researchers record outcomes such as knockdown, mortality, or changes in activity over a defined period compared with an untreated control. These assays help characterize dose-dependent toxicity, compare insecticides, identify promising larvicidal or adulticidal compounds, and investigate resistance in mosquito populations. The results support safer, more effective approaches to reducing disease transmission and managing insecticide resistance.

Protocols are described for assessing the toxicity of chemistries to immature and adult mosquitoes for development as new classes of larvicides, adulticides and endectocides. The protocols enable high-throughput testing of multiple chemistries at single-point dose and subsequent evaluation via dose response assay to determine toxicity on contact or via ingestion.

Reverse genetic approaches have proven extremely useful for determining which genes underly resistance to vector pathogens in mosquitoes. This video protocol illustrates a method used by the Dimopoulos lab to inject dsRNA into Anopheles gambiae mosquitoes, which harbor the malaria parasite. The technique manipulating the injection setup and injecting dsRNA into the thorax is illustrated.

C. elegans Survival Assay: A Short-Term Toxicity Test

2023

This video introduces a short-term survival assay that tests a treatment for acute toxicity or tests a genetic strain for resistance to a toxic condition. The example protocol shows an example survival assay in which worms are tested for their ability to resist oxidative stress induced by paraquat (PQ).
