Nadh Production

NADH production is the biological generation of reduced nicotinamide adenine dinucleotide, an electron-carrying molecule that links nutrient breakdown to cellular energy conversion. During glycolysis, pyruvate oxidation, and the citric acid cycle, enzymes remove hydrogen or electrons from metabolic intermediates and transfer them to NAD+, forming NADH through reduction. NADH then delivers these high-energy electrons to the mitochondrial electron transport chain, where their transfer through protein complexes drives proton pumping and supports ATP synthesis through oxidative phosphorylation. Measuring or manipulating NADH production helps researchers assess cellular respiration, metabolic activity, redox balance, and changes associated with disease, exercise, and biotechnology.

NADH Fluorescence Imaging of Isolated Biventricular Working Rabbit Hearts

The objective is to monitor the mitochondrial redox state of isolated hearts within the context of physiologic preload and afterload pressures. A biventricular working rabbit heart model is presented. High spatiotemporal resolution fluorescence imaging of NADH is used to monitor the mitochondrial redox state of epicardial tissue.

Simultaneous Measurement of Superoxide/Hydrogen Peroxide and NADH Production by Flavin-containing Mitochondrial Dehydrogenases

Mitochondria contain several flavin-dependent enzymes that can produce reactive oxygen species (ROS). Monitoring ROS release from individual sites in mitochondria is challenging due to unwanted side reactions. We present an easy, inexpensive method for direct assessment of native rates for ROS release using purified flavoenzymes and microplate fluorometry.

Here we describe a method to directly visualize microregional tissue hypoxia in the mouse cortex in vivo. It is based on concurrent two-photon imaging of nicotinamide adenine dinucleotide (NADH) and the cortical microcirculation. This method is useful for high resolution analysis of tissue oxygen supply.

A nicotinamide adenine dinucleotide (NADH)-coupled ATPase assay has been adapted to semihigh throughput screening of small molecule myosin inhibitors. This kinetic assay is run in a 384-well microplate format with total reaction volumes of only 20 µL per well. The platform should be applicable to virtually any ADP producing enzyme.
