Neutral Red

Neutral red is a phenazine dye used in chemistry as a pH indicator, biological stain, and analytical reagent. Its color changes from red in acidic conditions to yellow in alkaline conditions as protonation alters the dye’s electronic structure, while living cells can concentrate neutral red in acidic lysosomes. These properties support acid–base titrations, microscopy, cell-viability assays, and studies of membrane integrity. In particular, the neutral red uptake assay measures dye accumulation by viable cells, providing a practical readout for cytotoxicity and cellular health in chemical, pharmaceutical, and biomedical research.

Oil Red O Staining: A Technique for Staining Neutral Lipids in Hepatocytes for Detecting Hepatic Steatosis In Vitro

In this video, we demonstrate in vitro staining of neutral lipids in hepatocytes to detect hepatic steatosis or excessive fat accumulation in liver. Excessive fat is accumulated in the cells in form of lipid droplets. Oil Red O is a lipid soluble dye that binds to intracellular neutral lipids in these lipid droplets. When observed under a light microscope, these intracellular lipid droplets appear intensely red in color.

This study describes an imaging-based micro-neutralization assay to analyze the antigenic relationships between viruses. The protocol employs a flatbed scanner and has four steps, including titration, titration quantitation, neutralization, and neutralization quantitation. The assay works well with current circulating influenza A(H1N1)pdm09, A(H3N2), and B viruses.

An acid-base reaction is one in which a hydrogen ion, H+, is transferred from one chemical species to another. Such reactions are of central importance to numerous natural and technological processes, ranging from the chemical transformations within cells or lakes and oceans to the industrial-scale production of fertilizers, pharmaceuticals, and other substances essential to the society. There are several ways of defining an acid. In the context of aqueous solutions, an acid is a substance...
