Number Fraction Analysis

Number fraction analysis is a quantitative method that describes how many entities in a chemical population belong to a particular species, size class, or composition relative to the total number of entities. It works by classifying or measuring individual molecules, particles, or components, counting their occurrences, and dividing each count by the total population to obtain a normalized distribution. In chemistry, this approach helps characterize mixtures, polymers, colloids, aerosols, and other heterogeneous systems, while distinguishing numerical abundance from mass or volume fraction. Comparing these measures can reveal whether a sample contains many small components, fewer large components, or distinct subpopulations, supporting more accurate interpretation of composition and behavior.

Mitochondria play central roles in the regulation of metabolism and homeostasis. Subtle changes in mitochondrial metabolism that affect organismal physiology could be difficult to detect in whole organism metabolomics studies. Here we describe an isolation method that enhances the detection of subtle metabolic shifts in Drosophila melanogaster.

Polysome Fractionation and Analysis of Mammalian Translatomes on a Genome-wide Scale

Ribosomes play a central role in protein synthesis. Polyribosome (polysome) fractionation by sucrose density gradient centrifugation allows direct determination of translation efficiencies of individual mRNAs on a genome-wide scale. In addition, this method can be used for biochemical analysis of ribosome- and polysome-associated factors such as chaperones and signaling molecules.

This protocol separates insoluble cell wall and membrane proteins into simple fractions (1-5 proteins) using preparative isoelectric focusing (IEF) based on isoelectric point, followed by separation by molecular weight.
