Foam Cell Formation

Foam cell formation is the process by which macrophages and other vascular cells accumulate lipids, creating a foamy appearance that contributes to atherosclerotic plaque development. In arterial walls, modified low-density lipoprotein (LDL) is taken up through scavenger receptors, while disrupted cholesterol efflux promotes intracellular storage as lipid droplets and cholesteryl esters. These lipid-loaded cells release inflammatory mediators and can influence plaque growth and stability. Understanding foam cell formation helps clarify the cellular mechanisms underlying cardiovascular disease and supports research into lipid-lowering therapies, cholesterol transport, vascular inflammation, and strategies for preventing or treating atherosclerosis.

We describe a protocol to measure transmigration by monocytes across human endothelial monolayers and their subsequent maturation into foam cells. This provides a versatile method to assess the atherogenic properties of monocytes isolated from people with different disease conditions and to evaluate factors in blood which may enhance this propensity.

This study presents a methodology to prepare 3D, biodegradable, foam-like cell scaffolds based on biocompatible side-chain liquid crystal elastomers (LCEs). Confocal microscopy experiments show that foam-like LCEs allow for cell attachment, proliferation, and the spontaneous alignment of C2C12s myoblasts.

The tissue-specific extracellular matrix (ECM) is a key mediator of cell function. This article describes methods for synthesizing pure ECM-derived foams and microcarriers that are stable in culture without the need for chemical crosslinking for applications in advanced 3D in vitro cell culture models or as pro-regenerative bioscaffolds.
