Bone Morphogenic Protein 4

Bone morphogenic protein 4 (BMP4) is a secreted signaling protein in the transforming growth factor beta (TGF-β) superfamily that regulates cell fate, tissue patterning, and organ formation during embryonic development. After release, BMP4 binds type II and type I serine/threonine kinase receptors, triggering phosphorylation of SMAD1/5/8 and their partnership with SMAD4 to alter gene expression; extracellular antagonists can limit this signal. In developmental biology, BMP4 signaling helps establish body axes and influences differentiation in tissues including bone, heart, and nervous system, supporting research on congenital abnormalities, stem-cell differentiation, and tissue engineering.

The manuscript here provides a simple set of methods for analysing the secretion and diffusion of fluorescently tagged ligands in Xenopus. This provides a context for testing the ability of other proteins to modify ligand distribution and allowing experiments that may give insight into mechanisms regulating morphogen gradients.

The nature of the interactions between hematopoietic stem and progenitor cells (HSPCs) and bone marrow niches is poorly understood. Custom hardware modifications and a multi-step acquisition protocol allow the use of two-photon and confocal microscopy to image ex vivo labeled HSPCs homed within bone marrow areas, tracking interactions and movement.

Here, we present an overview of the preparation and animal handling procedures required to safely perform medical imaging in an animal biosafety level 4 laboratory. Computed tomography of a mock-infected guinea pig illustrates these procedures that may be used to evaluate the disease caused by a high consequence pathogen.

Murine Hind Limb Long Bone Dissection and Bone Marrow Isolation

Here we present a protocol for the dissection of hind limb long bones (femurs and tibiae) from the laboratory mouse. We further describe a rapid technique for bone marrow isolation from these bones that utilizes centrifugation for removal of bone marrow from the bone marrow space.
