Endothelial Network Formation

Endothelial network formation is the process by which endothelial cells organize into interconnected, vessel-like structures, providing a model for studying how vascular systems develop and function. During this process, growth factors such as vascular endothelial growth factor stimulate cell migration, proliferation, adhesion, and alignment within an extracellular matrix, allowing cells to connect and form branching networks. In developmental biology, these behaviors help explain vasculogenesis and angiogenesis during tissue growth. Network formation assays support research on vascular development, wound repair, tissue engineering, and diseases involving abnormal blood-vessel growth, while also enabling evaluation of pro- or anti-angiogenic treatments.

Endothelial Cell Tube Formation Assay for the In Vitro Study of Angiogenesis

The tube formation assay is a fast, quantifiable method for measuring in vitro angiogenesis. Endothelial cells are combined with conditioned media and plated on basement membrane extract. Tube formation occurs within hours and newly formed tubules easily quantified.

Primary human umbilical vein endothelial cells (HUVECs) were grown to confluence within a microfluidic network device. The endothelial cell junction and F-actin distributions were illustrated and the changes in intracellular calcium concentration and nitric oxide production in response to adenosine triphosphate (ATP) were quantified in real-time at individual cell levels.

This video demonstrates a technique for establishing a co-culture of interneurons with perivascular endothelial cells. The cells are added to a one-well insert placed inside a culture dish. After the cells adhere to the dish, the insert is removed, allowing the cells to proliferate and interact and form a co-culture.
