Germline Transplantation

Germline transplantation is a technique that transfers germline cells, such as primordial germ cells or germline stem cells, from a donor embryo into a recipient embryo to study how reproductive cells develop. The transplanted cells migrate to the recipient’s gonads, where they can proliferate, differentiate into sperm or eggs, and transmit donor-derived genetic information to offspring. In developmental biology, this approach separates the intrinsic properties of germ cells from the surrounding somatic environment and reveals how cellular signals guide germline development. It is also used to generate chimeric animals, preserve valuable genetic lines, and investigate fertility, inheritance, and reproductive disorders.

We present an immunofluorescence imaging-based method for spatial and temporal localization of active ERK in the dissected C. elegans gonad. The protocol described here can be adapted for visualization of any signaling or structural protein in the C. elegans gonad, provided a suitable antibody reagent is available.
