Mesenchyme Epithelium Interaction

Mesenchyme-epithelium interaction is the reciprocal communication between epithelial and mesenchymal tissues that coordinates tissue organization during development. Through paracrine signaling, extracellular matrix cues, and cell-cell adhesion, each tissue influences the other’s polarity, proliferation, migration, and differentiation; transitions between epithelial and mesenchymal states can further reshape developing structures. These interactions drive processes such as organ formation, branching morphogenesis, and the development of the kidney, lung, and teeth. Studying this signaling network helps explain how tissues acquire form and function, while also providing insight into congenital abnormalities, tissue regeneration, and pathological processes in which developmental programs become misregulated.

To unravel the earliest molecular mechanisms underlying prostate cancer initiation, novel and innovative human model systems and approaches are desperately needed. The potential of pre-prostatic urogenital sinus mesenchyme (UGSM) to induce pluripotent stem cell populations to form human prostate epithelium is a powerful experimental tool in prostate research.

Here, we present a protocol including mitochondrial tracing, direct co-culture procedures of mesenchymal stem cells (MSCs) and retinal pigment epithelial cells (ARPE19), as well as the methods for observing and statistically analyzing tunneling nanotubes (TNT) formation and mitochondrial transfer to characterize mitochondrial exchange via TNTs between MSCs and ARPE19 cells.

Electroporation of Craniofacial Mesenchyme

Craniofacial cartilages develop in close contact with other tissues and are difficult to manipulate in live animals. We are using electroporation to deliver molecular tools during growth of the craniofacial skeleton while bypassing early embryonic effects. This approach will allow us to efficiently test candidate molecules in vivo.
