Topography Imaging

Topography imaging is the measurement and visualization of a surface’s three-dimensional shape, including height variations, roughness, steps, and defects. It works by scanning or illuminating a surface with a probe, beam, or patterned light and converting position-dependent signals into a height map, often with nanometer- to micrometer-scale detail. In engineering, these images support the evaluation of machined parts, thin films, semiconductors, coatings, and biological or biomimetic materials. Quantitative topographic data helps researchers relate surface structure to friction, adhesion, wettability, optical behavior, and mechanical performance, guiding quality control, failure analysis, process optimization, and advanced device design.

Here, we provide the practical and technical steps for revealing global brain topography using a versatile wholemount immunohistochemical staining approach. We demonstrate the utility of the method using the well-understood cytoarchitecture and circuitry of cerebellum.

Laser Micromachining for Polymer Surface Topography Design

A method for imposing surface topography of light-absorbing composite polymer materials, particularly magnetoactive elastomers (MAEs), by using laser micromachining is presented. This method enables large flexibility in topographical designs and rapid prototyping. An example of structuring MAE surfaces, characterization, and their response to the external magnetic field is demonstrated.

Fast photochemical oxidation of proteins is an emerging technique for the structural characterization of proteins. Different solvent additives and ligands have varied hydroxyl radical scavenging properties. To compare the protein structure in different conditions, real-time compensation of hydroxyl radicals generated in the reaction is required to normalize reaction conditions.
