Antigen-adjuvant Emulsion

Antigen-adjuvant emulsion is a vaccine formulation that disperses an antigen and an immune-stimulating adjuvant within mixed oil and aqueous phases to strengthen protective immune responses. Its microscopic droplets influence antigen delivery, promote local innate immune activation, and enhance recruitment and activation of antigen-presenting cells, which process the antigen and stimulate adaptive immunity. In immunology and infection research, these emulsions help evaluate how formulation properties affect antibody production, T-cell responses, and vaccine durability. They are used to improve vaccine potency while guiding the design of safer, more effective immunization strategies against infectious diseases.

To induce experimental autoimmune encephalomyelitis, an animal model of multiple sclerosis, mice are immunized with a water-in-oil emulsion containing an autoantigen and complete Freund's adjuvant. While several protocols exist for the preparation of these emulsions, a rapid, simple, and standardized homogenization protocol for emulsion preparation is presented here.
