Cytokine Release Assay

A cytokine release assay is an immunological method that measures cytokines secreted by immune cells after exposure to a specific stimulus, helping characterize cellular activation and inflammatory responses. In a typical assay, blood-derived or cultured immune cells encounter an antigen, pathogen-associated component, or therapeutic compound, and released cytokines such as interferons, interleukins, or tumor necrosis factor are quantified using immunoassays or multiplex detection platforms. These measurements help assess host responses to infection, identify antigen-specific immunity, evaluate vaccine responses, and investigate drug-related immune activation. Cytokine release assays therefore connect cellular mechanisms with clinically relevant patterns of inflammation and immune regulation.

We describe a simple immunoassay to measure the production of pro-inflammatory cytokines, such as IL-1 beta production, in patients presenting with autoinflammatory phenotypes. By activating cells in whole blood cultures with pathogen-associated molecular patterns, specifically with lipopolysaccharide, cytokine secretion can be conveniently evaluated in whole blood supernatants.
