Ex Vivo Wound Model

An ex vivo wound model is a laboratory system that uses excised tissue to reproduce key features of wound repair and host defense outside the body, enabling controlled study of immunology and infection. Researchers create a standardized wound in viable tissue and maintain it under controlled culture conditions, where resident cells, extracellular matrix, immune signaling, and microbial interactions can be monitored over time. These models help evaluate inflammatory responses, pathogen growth, antimicrobial treatments, and tissue-repair processes while reducing reliance on whole-animal experiments. By bridging simplified cell cultures and complex in vivo studies, ex vivo wound models provide biologically relevant platforms for investigating infection mechanisms and testing therapeutic strategies.

Here we describe a novel diabetic murine model utilizing hairless mice for real-time, non-invasive, monitoring of biofilm wound infections of bioluminescent Pseudomonas aeruginosa. This method can be adapted to evaluate infection of other bacterial species and genetically modified microorganisms, including multi-species biofilms, and test the efficacy of antibiofilm strategies.
