Immune Cell Heterogeneity

Immune cell heterogeneity is the variation in immune cell types, states, functions, and responses within a population, a feature that shapes health, infection, and disease. It arises from differences in lineage, receptor expression, tissue environment, activation history, and exposure to cytokines or microbial signals, causing cells to respond differently to the same stimulus. In immunology and infection research, characterizing this diversity helps distinguish protective from harmful immune responses, explain pathogen persistence and inflammation, and identify cell populations associated with disease outcomes. Methods such as flow cytometry, single-cell sequencing, and functional assays can resolve these differences, improving biomarker discovery, vaccine design, and targeted therapies.

This method for isolating functional immune cells from the heart provides an alternative to the conventional methods of collagenase digestion, which causes unwanted immune cell activation, resulting in a decreased responsiveness of these cells. Our method of isolation yields functional cardiac immune cells by avoiding problems associated with enzymatic digestion.

Herein we describe two approaches to characterize cell polarization events in B lymphocytes during the formation of an IS. The first, involves quantification of organelle recruitment and cytoskeleton rearrangements at the synaptic membrane. The second is a biochemical approach, to characterize changes in composition of the centrosome, which undergoes polarization to the immune synapse.
