In Vitro Analysis

In vitro analysis is the study of biological processes outside a living organism, using isolated cells, tissues, fluids, or microorganisms under controlled laboratory conditions. Researchers manipulate defined variables in a culture or assay system, then measure outcomes through methods such as microscopy, immunoassays, molecular analysis, or growth-based testing. In immunology and infection research, these approaches help characterize immune-cell activation, pathogen replication, host–pathogen interactions, and the effects of drugs, antibodies, or other interventions. By enabling controlled comparisons and detailed measurements, in vitro analysis supports mechanism-focused research while providing evidence for experiments in more complex biological models.

We demonstrate a method to generate 3D breast cancer surrogates, which can be cultured using a perfusion bioreactor system to deliver oxygen and nutrients. Following growth, surrogates are fixed and processed to paraffin for evaluation of parameters of interest. The evaluation of one such parameter, cell density, is explained.

This procedure was established to be used for developing advanced 3D hepatic cultures in vitro, which can provide a more physiologically relevant assessment of the genotoxic hazards associated with nanomaterial exposures over both an acute or long-term, repeated dose regimes.
