Mediation Analysis

Mediation Analysis is a statistical framework for testing whether the effect of an exposure on an outcome operates through an intermediate variable, helping distinguish direct and indirect pathways. In immunology and infection research, it can examine how immune cell activation, cytokine signaling, or pathogen burden links an intervention or exposure to clinical or biological outcomes. The method estimates the total effect and partitions it into a direct effect and an indirect effect transmitted through the proposed mediator, typically using regression or related causal models. Careful study design and adjustment for confounding are essential because mediation results can clarify mechanisms, guide biomarker selection, and identify potential targets for vaccines, therapeutics, or infection control.

The Upf proteins that carry out nonsense-mediated decay (NMD) are found in all eukaryotic organisms, including humans. Each protein has an individual role, but they need to work in collaboration.
