Phenotypic Profile Analysis

Phenotypic profile analysis is a method for characterizing cells, organisms, or clinical samples by measuring observable features across multiple biological dimensions. It typically combines standardized experimental conditions with imaging, flow cytometry, molecular assays, or other high-content measurements, then compares multidimensional patterns to reference profiles or treatment responses. In immunology and infection research, this approach can distinguish immune-cell states, reveal how pathogens alter host phenotypes, and identify cellular responses to vaccines or antimicrobial compounds. By linking complex phenotype patterns to biological mechanisms, phenotypic profile analysis supports disease classification, biomarker discovery, therapeutic development, and evaluation of host-pathogen interactions.

Eukaryotic Polyribosome Profile Analysis

This article describes a protocol for the extraction of translating ribosomes from eukaryotic cells. Once extracted, ribosomes are separated into monosomes and polyribosomes by sucrose gradient fractionation to allow different ribosomal populations to be analyzed. As such, this method is the gold standard for examining the regulation of translation.

Automated Analysis of Intracellular Phenotypes of Salmonella Using ImageJ

Salmonella invades and replicates inside intestinal epithelial cells both in Salmonella-specific vacuoles and free in the cytosol (hyper-replication). A high-throughput fluorescence microscopy-based protocol is described here to quantify the intracellular phenotypes of Salmonella by two complementary image analyses through ImageJ, reaching single-cell resolution and scoring.

Metabolic Profile Analysis of Zebrafish Embryos

Zebrafish represent a powerful vertebrate model that has been under-utilised for metabolic studies. Here we describe a rapid way to measure the in vivo metabolic profile of developing zebrafish that allows the comparison of different mitochondrial function parameters between genetically or pharmacologically manipulated embryos, thereby increasing the applicability of this organism.

We illustrate the application of 1H(15N,αγ)12C resonant nuclear reaction analysis (NRA) to quantitatively evaluate the density of hydrogen atoms on the surface, in the volume, and at an interfacial layer of solid materials. The near-surface hydrogen depth profiling of a Pd(110) single crystal and of SiO2/Si(100) stacks is described.
