Spectral Cytometry

Spectral cytometry is a flow cytometry method that measures the complete fluorescence emission profile of labeled cells, enabling detailed multiparameter analysis of cellular identity and function. Lasers excite fluorescent dyes, while detectors capture their emitted light across a broad wavelength range; computational spectral unmixing then separates overlapping signatures using reference spectra and autofluorescence correction. In immunology and infection research, spectral cytometry supports simultaneous profiling of immune-cell subsets, activation states, cytokine responses, and pathogen-associated changes in complex samples. Its expanded fluorescence resolution can improve panel design and reveal rare or phenotypically distinct populations that conventional flow cytometry may miss.

This article describes spectral cytometry, a new approach in flow cytometry that uses the shapes of emission spectra to distinguish fluorochromes. An algorithm replaces compensations and can treat auto-fluorescence as an independent parameter. This new approach allows for the proper analysis of cells isolated from solid organs.

Photoacoustic ophthalmology (PAOM), an optical-absorption-based imaging modality, provides the complementary evaluation of the retina to the currently available ophthalmic imaging technologies. We report the using of PAOM integrated with spectral-domain optical coherence tomography (SD-OCT) for simultaneous multimodal retinal imaging in rats.
