Tnp-polysaccharide Immunization

TNP-polysaccharide immunization is an experimental approach that uses a trinitrophenyl (TNP) hapten linked to a repetitive polysaccharide to study antibody responses without conventional T-cell help. Repeating TNP groups extensively cross-link B-cell receptors, activating mainly marginal-zone and B-1 B cells through a T-independent mechanism and typically promoting rapid IgM production with limited immunological memory. This model helps researchers examine B-cell activation, antibody class switching, and innate-like humoral immunity, while providing a controlled system for investigating responses to carbohydrate-rich microbial antigens and evaluating how polysaccharide vaccines generate protective antibodies.

This study compared the polysaccharide contents of Hippophae rhamnoides subsp. sinensis Rousi and Hippophae gyantsensis (Rousi) Y. S. Lian obtained through hot water extraction and quantified through phenol-sulfuric acid colorimetry. We also investigated in vitro antioxidant activities of polysaccharides from these two Hippophae species through 2,2-diphenyl-1-picrylhydrazyl free radical scavenging experiments.
