Virulence Factors

Virulence factors are microbial structures, products, or strategies that enable a pathogen to colonize a host, evade defenses, cause tissue damage, or obtain nutrients, thereby influencing disease severity. They work through mechanisms such as adhesin-mediated attachment, capsule-based protection from phagocytosis, toxin production, secretion of effector proteins, and enzymes that promote invasion or disrupt host cells. In immunology and infection research, identifying these factors clarifies how pathogens interact with innate and adaptive immunity and supports the development of vaccines, anti-virulence therapies, diagnostic markers, and strategies for predicting clinical outcomes.

The method described here is used to induce the apoptotic signaling cascade at defined steps in order to dissect the activity of an anti-apoptotic bacterial effector protein. This method can also be used for inducible expression of pro-apoptotic or toxic proteins, or for dissecting interference with other signaling pathways.

Following in Real Time the Impact of Pneumococcal Virulence Factors in an Acute Mouse Pneumonia Model Using Bioluminescent Bacteria

Streptococcus pneumoniae is the leading pathogen causing severe community-acquired pneumonia and responsible for over 2 million deaths worldwide. The impact of bacterial factors implicated in fitness or virulence can be monitored in real-time in an acute mouse pneumonia or bacteremia model using bioluminescent bacteria.

This article illustrates the use of pulse-chase radio labeling in combination with site-specific photocrosslinking to monitor interactions between a protein of interest and other factors in E. coli. Unlike traditional chemical cross-linking methods, this approach generates high resolution “snapshots” of an ordered assembly pathway in a living cell.

Assessment of Leishmania Virulence within Cultured Macrophages

This video investigates Leishmania's virulence by treating macrophages with metacyclic promastigotes, resulting in the formation of intracellular amastigotes. The confirmation of Leishmania's virulence is achieved through nucleic acid fluorescent dye staining. This insight can contribute to the development of new therapies for leishmaniasis.

Source: Chen, Y., et al. Evaluating Virulence and Pathogenesis of Aeromonas Infection in a Caenorhabditis elegans Model. J. Vis. Exp. (2018).This video demonstrates the C. elegans Liquid Toxicity Assay with Aeromonas, a method used to evaluate bacterial virulence by measuring worm survival in a nutrient-limited liquid environment. The assay reveals strain-specific pathogenicity based on the extent of toxin-induced damage and mortality in C. elegans.
