Cisplatin Treatment

Cisplatin treatment is a chemotherapy approach that uses the platinum-based drug cisplatin to destroy or inhibit rapidly dividing cancer cells. After entering cells, cisplatin forms covalent bonds with DNA, creating intra- and interstrand crosslinks that distort the DNA structure, block replication and transcription, and trigger cell death. Clinicians use it alone or with other therapies for cancers including testicular, ovarian, bladder, lung, and head and neck tumors. Its effectiveness must be balanced with potential toxicities, particularly kidney damage, hearing loss, nerve injury, and suppression of blood-cell production, making dose adjustment, hydration, and patient monitoring essential in medical practice.

Electrochemotherapy is the local treatment of solid tumors. The main mechanism is electroporation-mediated permeabilization of the tumor cells' membrane which enables increased internalization of cytostatics like bleomycin and cisplatin. Thus, antitumor effectiveness is potentiated at the site of the application of electric pulses to the tumors.

This protocol describes the procedures to induce Acute Kidney Injury (AKI) in adult zebrafish using cisplatin as a nephrotoxic agent. We detailed the steps to evaluate the reproducibility of the technique and two techniques to analyze inflammation and cell death in the renal tissue, flow cytometry and TUNEL, respectively.
