Dextran Microinjection

Dextran microinjection is a technique that delivers dextran, often fluorescently labeled, directly into individual cells or tissues to track intracellular distribution and cell behavior. A fine micropipette applies controlled pressure to introduce the dextran solution across the cell membrane, while the molecule’s size and the membrane’s integrity influence its movement and retention. In medicine and biomedical research, this approach supports cell labeling, lineage tracing, studies of membrane permeability, and analysis of intracellular transport. By providing a visible, experimentally controlled tracer, dextran microinjection helps researchers examine cellular structure, connectivity, and responses in living biological systems.

In this protocol, we describe the direct cytoplasmic microinjection of cytochrome c protein into fibroblasts and primary sympathetic neurons. This technique allows for the introduction of cytochrome c protein into the cytoplasm of cells and mimics the release of cytochrome c from mitochondria, which occurs during apoptosis.

Zebrafish Microinjection Techniques

2023

One of the major advantages to working with zebrafish (Danio rerio) is that their genetics can be easily manipulated by microinjection of early stage embryos. Using this technique, solutions containing genetic material or knockdown constructs are delivered into the blastomeres: the embryonic cells sitting atop the yolk of the newly fertilized egg. Delivery into the cytoplasm is achieved either through direct injection into the blastomere, or via natural cytoplasmic movements that occur after a...

The objective of this protocol is to incorporate SDF-1α, a stem cell homing factor, into dextran sulfate-chitosan nanoparticles. The resultant particles are measured for their size and zeta potential, as well as the content, activity, and in vitro release rate of SDF-1α from the nanoparticles.
