Endothelial Cell Density

Endothelial cell density is the number of endothelial cells occupying a defined tissue area, a measure of how effectively this cell layer can maintain vascular or tissue barrier function. In medicine, it is commonly assessed in the cornea by microscopy or specular imaging, which counts cells and analyzes their size and shape; cell loss can reduce pump activity and disturb fluid balance. Clinicians use endothelial cell density to evaluate corneal health, monitor aging, disease, surgery, and medication effects, and estimate the risk of edema or vision-threatening decompensation. These measurements also support treatment planning and longitudinal patient monitoring.

We provide a method for isolating and culturing pure populations of heart valve endothelial cells (VEC). VEC can be isolated from either side of the cusp or leaflet and immediately following, underlying interstitial cell (VIC) isolation is straightforward.

This video presents the isolation of microvessels from a rat brain using a combination of density gradient centrifugation and enzymatic digestion mehods. The isolated microvessels can be cultured in a culture flask pre-coated with collagen and fibronectin matrix to obtain endothelial cell colonies.

Hematopoietic stem and progenitor cells (HSPC) derive from specialized (hemogenic) endothelial cells during development, yet little is known about the process by which some endothelial cells specify to become blood forming. We demonstrate a flow-cytometry based method allowing simultaneous isolation of hemogenic endothelial cells and HSPC from murine embryonic tissues.
