Intravascular Imaging

Intravascular imaging is a group of catheter-based methods that visualize the inside of blood vessels, providing information that angiography alone may not reveal. During a procedure, a miniature ultrasound transducer or optical coherence tomography (OCT) probe travels through the vessel and generates cross-sectional images of the lumen, vessel wall, plaque, and implanted devices. In medicine, these techniques help characterize atherosclerotic lesions, assess the severity and composition of coronary disease, and guide stent placement by identifying problems such as incomplete expansion or malapposition. Intravascular imaging therefore supports more precise diagnosis, treatment planning, and evaluation of outcomes in vascular interventions.

We detail a new near-infrared fluorescence (NIRF) catheter for 2-dimensional intravascular molecular imaging of plaque biology in vivo. Here, we present a protocol for intravital imaging of an attenuated BSL-2-approved Mycobacterium tuberculosis. strain in the murine lung using a permanent lung window. This approach enables longitudinal, single-cell-resolution analysis of early host-pathogen interactions, including bacillary localization, vascular dynamics, and macrophage uptake within the same lung microenvironment.

Here, we describe a simple method of intracerebroventricular and intravascular injection of viral particles or fluorescent microbeads into the neonatal mouse brain. The localization pattern of the virus and nanoparticles could be detected by microscopic evaluation or by in situ hybridization.
