Msc Hydrogels

MSC hydrogels are hydrated, three-dimensional polymer networks designed to encapsulate or support mesenchymal stem/stromal cells (MSCs) for biomedical research and therapy. Their water-rich matrix provides a tissue-like environment, while polymer crosslinking controls stiffness, porosity, degradation, cell retention, and the diffusion of nutrients or signaling molecules; these properties can influence MSC survival and differentiation. In medicine, MSC hydrogels are investigated as injectable or implantable platforms for tissue repair, regenerative medicine, drug delivery, and cell-based therapies. By combining cellular activity with tunable biomaterials, they may improve localized treatment and help model how cells respond to engineered tissue environments.

Here, a protocol (MitoCeption) is presented to transfer mitochondria, isolated from human mesenchymal stem cells (MSC), to glioblastoma stem cells (GSC), with the goal of studying their biological effects on GSC metabolism and functions. A similar protocol can be adapted to transfer mitochondria between other cell types.

2023

Collagen is another widely used biomaterial that has found popularity in commercial applications, such as photography. Collagen has more recently been used in tissue engineering applications, by creating hydrogels that provide structure to engineered tissue. This video introduces collagen as a biomaterial, demonstrates how it is harvested from porcine skin, and shows how the material is used to create a hydrogel for tissue engineering applications. Finally, several applications of the material...

Sparks, Department of Materials Science and Engineering, The University of Utah, Salt Lake City, UT Hydrogels are a versatile class of cross-linked polymers produced through relatively simple procedures and with generally inexpensive materials. They can be formed from solution and involve a polymer backbone formed from monomer reagents, an initiator which makes the polymer reactive and a crosslinking species which binds the polymer chains...

Production and Administration of Therapeutic Mesenchymal Stem/Stromal Cell (MSC) Spheroids Primed in 3-D Cultures Under Xeno-free Conditions

The therapeutic potential of mesenchymal stem/stromal cells (MSCs) is well-documented, however the best method of preparing the cells for patients remains controversial. Herein, we communicate protocols to efficiently generate and administer therapeutic spherical aggregates or 'spheroids' of MSCs primed under xeno-free conditions for experimental and clinical applications.
