Nude Mice Models

Nude mice models are immunodeficient laboratory animals, typically athymic mice with a congenital absence or severe impairment of the thymus, used to study disease and treatment in vivo. Their reduced T-cell development limits cellular immune rejection, allowing transplanted human tumor cells, tissues, or other grafts to survive and grow under controlled conditions. In medicine, these xenograft models support cancer biology, therapeutic screening, tumor growth assessment, and investigation of treatment responses. Because they do not reproduce a complete human immune system, findings require careful interpretation and may be complemented by humanized or other immune-competent models.

Mycobacterium leprae, the causative agent of leprosy, does not grow in vitro. We describe an easy to follow protocol to prepare a bacillary suspension to ensure the maintenance of large quantities of M. leprae for a variety of applications. Protocols for propagation by mouse footpad inoculation, evaluation of viability, freezing and thawing bacillary stock are described in detail.

Fat grafting is an essential technique for reconstructing soft tissue deficits. However, it remains an unpredictable procedure characterized by variable graft survival. Our goal was to devise a mouse model that utilizes a novel imaging method to compare volume retention between differing techniques of fat graft preparation and delivery.

Multi-modal Imaging of Angiogenesis in a Nude Rat Model of Breast Cancer Bone Metastasis Using Magnetic Resonance Imaging, Volumetric Computed Tomography and Ultrasound

In the pathogenesis of bone metastasis, angiogenesis is a crucial process and therefore represents a target for imaging and therapy. Here, we present a rat model of site-specific breast cancer bone metastasis and describe strategies to non-invasively image angiogenesis in vivo using magnetic resonance imaging, volumetric computed tomography and ultrasound.
