3d Co-culture Spheres

3D co-culture spheres are three-dimensional cellular aggregates formed by growing multiple cell types together, providing a model of the cell-cell interactions that shape neural tissue. Under conditions that limit attachment to a flat surface, cells cluster through cell adhesion and extracellular signaling, generating a compact structure with spatial organization and diffusion gradients. In neuroscience, these spheres can combine neurons with glial or other support cells to examine communication, neurite development, and responses to injury or treatment. Their three-dimensional architecture offers a more physiologically relevant setting than conventional two-dimensional cultures, supporting studies of neural development, disease mechanisms, and potential therapeutics.

Here, we describe how to produce, expand, and immunolabel postnatal hippocampal neural progenitor cells (NPCs) in three-dimensional (3D) culture. Next, using hybrid visualization technologies, we demonstrate how digital images of immunolabelled cryosections can be used to reconstruct and map the spatial position of immunopositive cells throughout the entire 3D neurosphere.

2023

Source: Jose Roberto Moreto and Xiaofeng Liu, Department of Aerospace Engineering, San Diego State University, San Diego, CA Wind tunnel tests are useful in the design of vehicles and structures that are subjected to airflow during their use. Wind tunnel data are generated by applying a controlled air flow to a model of the object being studied. The test model usually has a similar geometry but is a smaller scale compared to the full-sized object. To ensure accurate and useful data is collected...
