Stereotaxic Infusion

Stereotaxic infusion is a neuroscience technique that delivers drugs, tracers, genetic materials, or other solutions to precise locations within the brain or spinal cord. Using a stereotaxic apparatus, researchers identify target coordinates relative to anatomical landmarks, guide a needle or cannula through the tissue, and control the infusion volume and rate to limit spread beyond the intended region. This approach supports localized manipulation of neural circuits, measurement of behavioral or physiological responses, and investigation of brain function, disease mechanisms, and therapeutic strategies. It is especially valuable when experiments require spatially selective delivery that systemic administration cannot provide.

Here, we present a protocol for direct stereotaxic brain infusion of amyloid-beta. This methodology provides an alternative in vivo mouse model to address the short-term effects of amyloid-beta on brain neurons.

Direct Stereotaxic Brain Infusion of Amyloid-Beta Proteins for an Alzheimer's Disease Model

This video demonstrates the precise stereotaxic infusion of amyloid beta into the dentate gyrus to establish a mouse model of Alzheimer's disease. Accurate coordinate determination and controlled delivery of amyloid beta proteins result in neuronal degradation, disrupting memory processing and contributing to Alzheimer's disease development.

The monitoring of extracellular neurotransmitter levels in distinct brain regions of freely moving animals offers insights on the link between neurotransmitter release and behavior. In vivo microdialysis coupled with electrochemical detection provides excellent anatomical and chemical resolution; and information on how basal neurotransmission is altered by pharmacological or physiological manipulations.

Rodent Stereotaxic Surgery

Stereotaxic (or stereotactic) surgery is a method used to manipulate the brain of living animals. This technique allows researchers to accurately target deep structures within the brain through the use of a stereotaxic atlas, which provides the 3D coordinates of each area with respect to anatomical landmarks on the skull. After the skull is exposed, anesthetized animals are mounted on a specialized instrument known as a stereotaxic frame, which enables the precise placement of experimental...
