Synaptic Bouton Visualization

Synaptic bouton visualization is an imaging-based approach used to identify and characterize the small presynaptic terminals where neurons release neurotransmitters, making it important for linking synaptic structure to neural communication. Fluorescent proteins, dyes, or antibody-based markers label bouton components, while light or electron microscopy resolves their location, size, shape, and relationship to postsynaptic structures. In neuroscience, these measurements help quantify synapse density, track bouton formation and elimination, and assess structural changes after neuronal activity, development, or disease. Combined with live-cell imaging and functional indicators, the method connects presynaptic morphology with neurotransmitter release and supports studies of circuit plasticity and neurological disorders.

Recording Synaptic Currents at the Drosophila Larval Neuromuscular Junction

2025

This video demonstrates the technique for recording synaptic currents from a synaptic bouton at the Drosophila larva's neuromuscular junction. This technique enables monitoring the activity of a single synaptic bouton, which is important for the study of mutant fly lines with enhanced spontaneous or asynchronous synaptic activity.

FM dyes have been of invaluable help in the understanding of synaptic dynamics. FMs are normally followed under the fluorescent microscope during different stimulation conditions. However, photoconversion of FM dyes combined with electron microscopy allows the visualization of distinct synaptic vesicle pools, among other ultrastructure components, in synaptic boutons.
