Fibrosis Model

A fibrosis model is an experimental or computational representation of fibrosis, the pathological accumulation and remodeling of connective tissue that can impair organ structure and function. In physics-based research, these models examine how extracellular-matrix composition, tissue stiffness, mechanical stress, and force transmission influence the behavior of cells and developing scar tissue. Models may combine measurements of material properties with mathematical descriptions of deformation, transport, or cell–matrix interactions. They support quantitative analysis of how local physical conditions contribute to disease progression and help evaluate potential strategies for controlling tissue mechanics in organs affected by fibrosis.

We describe a method to produce an animal model of liver fibrosis in the rat, and assess the degree of fibrosis by histological examination of the liver. The model can be used to study the development of liver disease as well as to test the efficacy of potential anti-fibrotic agents.

Mouse model of metabolic dysfunction-associated steatotic liver disease (MASLD) with metabolic dysfunction, hepatic gene expression changes, and liver histopathological alterations that resemble human MASLD, including fibrosis that progresses to advanced fibrosis stage 3. This model can be used in studies of MASLD pathophysiology and in pre-clinical studies of new therapies.

This protocol demonstrates how the Src homology 2 domain-containing 5'-inositol phosphatase (SHIP)-deficient mouse model of Crohn disease (CD)-like ileal inflammation and fibrosis can be used to test novel therapeutics for CD.

A Refined Aerosol-Based Intratracheal Bleomycin Delivery Method for Reproducible and Minimally Invasive Mouse Models of Pulmonary Fibrosis

Here, we present a protocol to model pulmonary fibrosis in mice using aerosolized bleomycin delivered via the trachea. This optimized method enables uniform, quantitative, and precise delivery without surgical incision, enhancing the reproducibility and reliability of bleomycin-induced fibrosis.

Ischemia-reperfusion Model of Acute Kidney Injury and Post Injury Fibrosis in Mice

We describe models of moderate and severe ischemia-reperfusion-induced kidney injury in which the mice undergo unilateral renal pedicle clamping followed by simultaneous or delayed contralateral nephrectomy, respectively. These models consistently give rise to renal dysfunction and post-injury fibrosis, but injury severity and survival are dependent on mouse background, age and surgical equipment.
