Neuronal Axons

Neuronal axons are specialized extensions of nerve cells that transmit electrical signals from the cell body to other neurons, muscles, or glands, making rapid communication throughout the nervous system possible. An action potential travels along the axon when changes in membrane potential open voltage-gated ion channels, while myelin can insulate the axon and increase conduction speed through saltatory conduction between nodes of Ranvier. Axon terminals convert these electrical signals into chemical messages by releasing neurotransmitters across synapses. Studying axonal development, signaling, injury, and regeneration helps explain nervous system function and supports research into neurological disorders and potential repair strategies.

Axons are long, cytoplasmic processes of nerve cells capable of propagating electrical impulses known as action potentials. The cytoplasm or axoplasm of an axon contains neurofibrils, neurotubules, small vesicles, lysosomes, mitochondria, and various enzymes, all encased within the axolemma, the plasma membrane of the axon. The axon attaches to the cell body at a cone-shaped elevation called the axon hillock. The initial part of the axon, closest to the hillock, is known as the initial segment.

Inducing Axonal Varicosities with a Micromechanical Stimulus on Neurons

This video demonstrates the induction of axonal varicosities by applying micromechanical stimulus on neurons. The system comprises a micropipette attached to a syringe containing buffer via tubing. Using a micromanipulator, the micropipette is positioned above the neurons. Fluid pressure is then applied to induce varicosities in axons.

This work details a standard immunohistochemistry method to visualize motor neuron projections of late stage-16 Drosophila melanogaster embryos. The filleted preparation of fixed embryos stained with FasII antibody provides a powerful tool to characterize the genes required for motor axon pathfinding and target recognition during neural development.
