Trypan Blue Quenching

Trypan blue quenching is a fluorescence-based technique that suppresses signal from antibody or dye molecules remaining outside cells, enabling measurement of intracellular or internalized material. Trypan blue enters damaged cells but generally remains outside intact cells, where it absorbs or quenches accessible fluorophores without substantially affecting protected fluorescence within viable cells. In immunology and infection research, this approach helps distinguish surface-bound from internalized antibodies, receptors, pathogens, or pathogen-derived components in microscopy and cell-based assays. By improving interpretation of uptake and localization measurements, Trypan blue quenching supports studies of cellular entry, immune recognition, phagocytosis, and host-pathogen interactions.

Quorum-quenching enzymes are anti-virulent and anti-bacterial options that can mitigate pathogenesis without risk of incurring resistance, by preventing the expression of virulence factors and genes associated with antibiotic resistance and biofilm formation. In this study, we report a method that demonstrates the efficacy of quorum-quenching enzymes in bacterial biofilm disruption.

Cerebral Blood Oxygenation Measurement Based on Oxygen-dependent Quenching of Phosphorescence

We present an experimental procedure for measuring the partial pressure of oxygen (pO2) in cerebral vasculature based on oxygen-dependent quenching of phosphorescence. Animal preparation and imaging procedures were outlined for both large field of view CCD-based imaging of pO2 in rats and 2-photon excitation based imaging of pO2 in mice.

The preparation and exfoliation of CaCuSi4O10 and BaCuSi4O10 are described. Upon stirring in hot water, CaCuSi4O10 spontaneously exfoliates into monolayers, whereas BaCuSi4O10 requires ultrasonication in organic solvents. Near infrared (NIR) imaging illustrates the NIR emission properties of these materials, and aqueous dispersions of these nanomaterials are useful for solution processing.

Biofunctionalized Prussian Blue Nanoparticles for Multimodal Molecular Imaging Applications

This protocol describes the synthesis of biofunctionalized Prussian blue nanoparticles and their use as multimodal, molecular imaging agents. The nanoparticles have a core-shell design where gadolinium or manganese ions within the nanoparticle core generate MRI contrast. The biofunctional shell contains fluorophores for fluorescence imaging and targeting ligands for molecular targeting.
