Macromolecular Crystallography

Macromolecular crystallography is a structural biology technique that determines the three-dimensional arrangements of atoms in proteins, nucleic acids, and their complexes, helping explain how these molecules function. In biochemistry, researchers purify and crystallize a macromolecule, expose the crystal to X-rays, and interpret the resulting diffraction pattern to calculate an electron-density map and build an atomic model. These structures reveal active sites, binding interactions, conformational changes, and molecular assemblies. The method supports structure-function studies, enzyme mechanism analysis, ligand optimization, and structure-guided drug design, making it a powerful approach for connecting molecular architecture with biological activity.

An All-in-one Sample Holder for Macromolecular X-ray Crystallography with Minimal Background Scattering

A novel sample holder for macromolecular X-ray crystallography along with a suitable handling protocol is presented. The system allows crystal growth, crystal soaking and in situ diffraction data collection at both, ambient and cryogenic temperature without the need of any crystal manipulation or mounting.

Optimization of Crystal Growth for Neutron Macromolecular Crystallography

Structural studies of biomacromolecules by crystallography require high-quality crystals. Here we demonstrate a protocol that can be used by OptiCrys (a fully automated instrument developed in our lab) and/or microdialysis buttons for growing large high-quality crystals based on knowledge of the crystallization phase diagram.

We present a protocol to obtain cell-derived matrices rich in extracellular matrix proteins, using macromolecular crowders (MMC). In addition, we present a protocol which incorporates MMC in 3D organotypic skin co-culture generation, which reduces culture time while maintaining maturity of construct.
