Substrate Oxidation Analysis

Substrate oxidation analysis is the study of how biological molecules are chemically oxidized to release energy or provide intermediates for metabolism. It examines redox reactions in which electrons or hydrogen atoms move from a substrate to an electron acceptor, often through enzyme-catalyzed pathways that generate reduced cofactors and, in aerobic systems, consume oxygen. In biochemistry, this approach helps characterize enzyme activity, compare the use of carbohydrates, lipids, and other substrates, and assess metabolic pathway function. Measurements of oxidation can reveal changes in cellular energy production, metabolic regulation, and substrate preference, supporting research in physiology, disease mechanisms, biotechnology, and drug development.

This video demonstrates a quantification technique for CO2 released during substrate oxidation using 14C-radiolabeled substrates. The released 14CO2 is trapped in an alkaline solution and quantified by a scintillation counter. The oxidation of different substrates varies between tissues and reflects the pathophysiological condition of the tissue.

Here we report a protocol to measure oxidative stress in living zebrafish embryos. This procedure allows reactive oxygen species (ROS) detection in both whole embryo tissues and single-cell populations. This protocol will accomplish both qualitative and quantitative analyses.

Aerosol-assisted Chemical Vapor Deposition of Metal Oxide Structures: Zinc Oxide Rods

Columnar zinc oxide structures in the form of rods are synthesized via aerosol-assisted chemical vapor deposition without the use of pre-deposited catalyst-seed particles. This method is scalable and compatible with various substrates based on either silicon, quartz or polymers.
