Ex Vivo Lung Specimens

Ex vivo lung specimens are lungs or lung sections maintained outside the body for controlled study, providing access to native tissue architecture while reducing the complexity of whole-animal experiments. In bioengineering research, researchers preserve specimens by supplying appropriate perfusion, ventilation, temperature, and nutrient conditions, allowing cells, extracellular matrix, airways, and blood vessels to remain structurally and functionally relevant for a limited period. These preparations support evaluation of biomaterials, tissue-engineered grafts, imaging methods, drug-delivery systems, and mechanical responses, while enabling direct measurement of transport, injury, and repair. Their use helps bridge in vitro models and in vivo studies and can improve the design of regenerative lung technologies.

We describe a relatively simple method for ex vivo live imaging of the tumor cell-stroma interactions within lung metastasis, utilizing fluorescent reporters in mice. Using spinning-disk confocal microscopy, this technique enables visualization of live cells for at least 4 hr and could be adapted to study other inflammatory lung conditions.

Establishment of an Ex Vivo Lung Perfusion Rat Model for Translational Insights in Lung Transplantation

Here, we describe the necessary steps for establishing a rat EVLP model and show the inflammatory profile associated with the perfused lungs. The aim is to propagate knowledge and experiences about the rat EVLP model, enabling the integral understanding of the biological responses associated with that revolutionary technique.

Ex-Vivo Lung Perfusion (EVLP) has allowed lung transplantation in humans to become more readily available by enabling the ability to assess organs and expand the donor pool.
