N-glycoproteomics

N-glycoproteomics is the comprehensive study of proteins carrying N-linked glycans, including the identity, structure, abundance, and attachment sites of these sugar modifications. In a typical workflow, glycoproteins or glycopeptides are enriched, N-glycans or intact glycopeptides are separated by liquid chromatography, and mass spectrometry measures their mass and fragmentation patterns; enzymatic treatment with PNGase F can aid site assignment. In biology, these data reveal how glycosylation regulates protein folding, stability, trafficking, and cell communication. The approach supports biomarker discovery, disease research, and characterization of therapeutic proteins while exposing glycan heterogeneity that conventional proteomics may miss.

This paper describes a methodology to prepare cardiovascular tissue samples for MS analysis that allows for (1) the analysis of ECM protein composition, (2) the identification of glycosylation sites, and (3) the compositional characterization of glycan forms. This methodology can be applied, with minor modifications, to the study of the ECM in other tissues.

The footprint identification technique, a robust, accurate and cost-effective image classification system, is a new approach to monitoring cheetahs.
